Description

Description: Vero/hSLAM was derived by transfection of Vero cells with a vector plasmid (pCXN2) containing the neomycin resistance gene and an expression plasmid (pCAG-hSLAM) encoding the human signaling lymphocytic activation molecule (SLAM) “, also known as CDw150, which is a receptor for measles virus. Clones of transfected cells were selected in the presence of geneticin (also known as G418). Vero/hSLAM cells can be used to isolate measles virus from human clinical samples and are a substitute for the B95a cell line for this purpose. Mumps susceptibility has also been demonstrated at the depositor's laboratory.

Growth Medium: MEM with Earle's salts + 2mM Glutamine + 5% Foetal Bovine Serum (FBS) + 0.4mg/ml Geneticin. It is essential that geneticin is added to the culture medium as it is required to maintain the expression of the SLAM gene.

Subculturing Procedure: Split sub-confluent cultures (70-80%) 1:3 to 1:4 i.e. seeding at 1-3×10,000 cells/cm² using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
