Description
Description: COS-7 cell line (ECACC catalogue no. 87021302) adapted to grow in the chemically defined protein- and peptide-free InVitrus medium (Cell Culture Technologies). This cell line is substrate dependent i.e. adherent. COS-7 was derived from CV-1 simian cells transformed by an origin-defective mutant of SV40.

Growth Medium: InVitrus medium (Cell Culture Technologies http://www.cellculture.com/) + 4mM Glutamine + 5 ug/ml Insulin. When freezing cells down use culture medium (50:50 conditioned medium:fresh medium) + 10% DMSO.
Subculturing Procedure: In order to prevent syncitia formation, do not allow cells to become confluent. Split semi-confluent cultures 1:3 i.e. seeding at 4×10,000 cells/cm² using an animal component free dissociation reagent; incubate at 37°C. Please note: Once cells have detached from the flask it is necessary to neutralise the dissociation reagent using a compatible inhibitor . Centrifuge the cells; 100-150 x g for 3-5 minutes; count and re-suspend at the recommended seeding density in fresh medium
