Description
Description: Overproduces ornithine decarboxylase in response to polyamine deprivation.

Growth Medium: EMEM (EBSS) + 2mM Glutamine + Ham’s F12 (without Putrescine) (1:1) + 0.1% BSA.
Subculturing Procedure: Split sub-confluent cultures (70-80%) 1:5 to 1:10 i.e. seeding at 5×1,000 to 1×10,000 cells/cm² using cold PBS and 0.25% cold trypsin. Place flasks at 4°C for 5-10 minutes or until cells detach. Culture cells at 37°C; 5% CO2. Pre-soak flasks in porcine collagen (Sigma G-2625) overnight at room temperature or 2hr at 37°C. Serum is only used to initially resuscitate from frozen and should be added prior to cryopreservation.
