Description

Description: Established from a transplantable pancreatic tumour of the islets of Langerhans. The tumour originated in an irradiated NEDH inhibited albino rat and was maintained by serial transplantation in Nedh rats. After transplantation into athymic nude mice the cell line RIN-m was established from the fourth nude mouse heterotransplant, 9 days after the tumour was removed cells were exposed to cystine-free medium resulting in the complete elimination of fibroblast-like cells. RIN-m cells secrete insulin and somatostatin. High levels of L-dopa-decarboxylase and membrane bound secretory granules were detected. RIN-5F (95090402) was established as a secondary clone from RIN-m.

Growth Medium: RPMI 1640 + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).

Subculturing Procedure: Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 1-3×10,000 cells/cm² using 0.05% trypsin or trypsin/EDTA; 5% CO2; 37°C. Cell growth is relatively slow, with the cells taking up to 10 days to achieve 70-80% confluence following resuscitation from frozen or after sub-culture. Media change the cultures every 2-3 days to encourage growth if not sub-culturing.
